Developing ML prediction models for the identification of adverse maternal and neonatal outcomes
Stillbirth is often referred to as a ‘silent epidemic’ accounting for the fifth leading global cause of deaths (when compared with causes of death in all ages)-outranking cancer, tuberculosis (TB) and HIV. That is: losing 5 precious babies each minute. The rate in Canada remains high at 8.9 per 1,000 births- which has not reduced in 20 years. Stillbirths including maternal death rates are significantly higher for First Nations and Inuit populations in Canada compared to non-Aboriginal populations and, in low- and middle-income countries (LMICs), mainly related to lack of health infrastructure and clinical monitoring during pregnancy. Despite of such socio-economic disparities most cases are associated with a missed or delayed diagnosis. Therefore, there is an urgent critical need to discover novel biomarkers (prognostic and predictive) that enable diagnosis of adverse gestational outcomes (APO) earlier.Evaluating the endometrial microenvironment stiffness and pre-eclampsia
Pregnancy is dependent on the transformation of the endometrium, a process driven by the differentiation of endometrial stromal cells (EnSCs) into specialized decidual cells. The decidua contributes to early nutrient exchange, production of cytokines, and growth factors as well as supports the development of new blood vessels, modulates extravillous trophoblast (EVT) invasion, and acts as a protective barrier against infections and maternal host immune responses.
Disturbances in the uterine microenvironment are associated with miscarriages and third-trimester perinatal morbidity and mortality caused by pre-eclampsia (PE) and intrauterine growth restriction, small-for-gestational age, pre-term birth, and stillbirth. It is increasingly evident that abnormal levels of inflammatory and growth/signaling factors produced in the decidua during early pregnancy play a decisive role in the deregulation of the local microenvironment leading to several disorders of pregnancy. Our research investigates how impaired cytoskeletal rearrangement and stiffness participates in the pathophysiology of pre-eclampsia.Evaluation of new point-of-care tools for use in pregnancy
Rapid and frequent screening of molecules is necessary for precise interventions in severe pathophysiological conditions. We are evaluating the use of point-of-care-ready bioanalytical platforms for screening of multiplexed-cytokines based on one-dimensional ion-sensitive field-effect transistors arrays (nanoISFETs) of silicon fabricated at wafer-scale via nanoimprint lithography in collaboration with RWTH, Aachen, Germany.GrantsPrincipal Applicant.
